The special train will stop at Borivali, Vasai Road, Panvel, Lonavala, Pune, Dhond, Solapur, Gundakal, Katpadi, Vellore Cantt, Thiruvannamalai, Villupuram, Cuddalore and Port.
The development will benefit pilgrims from Kerala.
A ticket can be bought for Rs 2,200 as part of the inaugural offer, and the tickets can be reserved individually or in groups.
With the introduction of the new train, pilgrims from Kerala can spend almost 12 hours on Sunday at Velankanni before boarding the return train.
Velankanni, a small hamlet at the shores of the Bay of Bengal, is home to one of the world's biggest Catholic pilgrimage centres.
